Samples of feces and sera obtained from 3-year-old children were studied to increase the knowledge about the circulations of virus vaccines during the massive campaigns. The use of the oral polio vaccine with schemes of massive campaigns allows the circulation of the virus vaccine 2 months after their completion. The use of continual vaccination schemes makes possible the circulation of the virus vaccine for longer periods of time. Even in populations with a low immunity coverage, epidemic outbreaks of the vaccine-derived virus may appear. The total of poliovirus vaccine isolated in 2-year-old children (11 cases, 11.0 %) and the boosts of neutralizing antibodies (51 cases, 51.0 %), show a contradiction between the verification of the infections caused by isolations of the viruses and the results of boosts. The low percentage of isolations of virus vaccine and the highly significant percentages of of seroconversions or boosts to polio virus, allow to infer the occurrence of silent circulation. The silent circulation self limited to 2 months after concluding the campaign is due, among other causes, to the homologous or not induced response by the primary infection with the first dose of oral polio vaccine and by the secondary infections. The self limitation of the circulation of the polio viruses in massive campaigns constitutes an excellent prevention of the risks represented by the vaccine-derived viruses appearing in vaccinations with continual schemes.

Viral infectivity of Hawaii, 3 Peri and Riberao Pretto dengue 1 strains was evaluated in Vero and HepG2 cell lines by indirect immunofluorescence techniques. Dengue virus cellular tropism in vitro is diverse. They may replicate themselves in a great variety of cellular cultures, whose sensibility to viral infection is variable. The greatest percentage of infected cells in the HepG2 cell line was obtained with the highest multiplicities of infection (0.04 for Hawaii and Riberao Pretto strains and 0.01 for 3 Perú). The highest percentage of infected HepG2 and Vero cells for the studied strains and the greatest titer in the viral overnadant was obtained on the 5th day. Vero cell line was more sensitive to viral infection, since for the same multiplicity values it was detected a higher number of fluorescent cells and a better viral titre in the line of this overnadant than in the HepG2. The best result was obtained with the Hawaii strain that allowed to confirm faster the infection of the studied cellular lines.

A methodology was developed for the selection of genetically modified strains of Vibrio cholerae 01 and 0139 aimed at obtaining oral attenuated candidate vaccines against cholera. The modified strains underwent microbiological characterization, bacterial susceptibility and different biological tests (mean lethal dose, colonizing capacity, adherence in mice, ligated intestine and intraduodenal inoculation in rabbits as virulence and potency tests. The strains 81, 638, 638T and 1333 were evaluated in clinical trials to determine their reactogenicity and immunogenicity. All the strains were sensitive to tetracycline and doxoclycine. They showed their attenuation and immunogenicity in animal models. The strains 638 and 1333 proved to be immunogenic and non reactogenic in volunteers.

The feces of 397 patients with acute diarrheal disease (ADD) and of other 121 patients without diarrea (control group) were studied in the state of Mérida, Venezuela, from June 1993 to December 1994. The genus Aeromonas was identified in patients with ADD in 11.83% and in 5.78% of the patients from the control group. On studying the virulence factors described for Aeromonas (enterotoxin, cytotoxin, hemaglutinins, cellular hydrofibrosity, and hemolytic activity) in the isolated strains, it was detected that all presented at least one of the factors investigated associated with enteropathogenicity. Of the isolated species, Aeromonas caviae was the most frequently identified. All these results suggest that the Aeromonas species are potential enteric pathogens in this population.

The wide symptomatic range gave origin to more than 50 different diagnoses in the studied patients, who were confirmed that they suffered from chronic leptospirosis by the diverse tests made.

A multi-serovar ELISA based on the outer membrane Lipoprotein L41 (LipL41) of pathogenic Leptospira was developed to increase sensitivity by using a single test antigen. A sensitivity of 99 % and a specifity of 92 % could be achieved. The established diagnostic polymerase chain reaction is also able to detect fast and reliably pathogenic Leptospira in different clinical samples.
