Few animal models can reproduce early metastasis. In addition, no model has been reported that reproduces cancer-related hypercoagulability, which is characteristic of pancreatic ductal adenocarcinoma and other adenocarcinomas. We hypothesized that the reason why the commonly used orthotopic cell xenograft model cannot reproduce the disease is inadequate construction of the cancer microenvironment. We developed an orthotopic tissue fragment xenograft model in which tumor tissue was transplanted into the pancreas of mice while preserving the microenvironment. Briefly, we injected single cancer cells subcutaneously to form a tumor, which was then cut with a scalpel into tumor fragments. A fragment was then sutured and fixed to the surface of the pancreatic tail. In this study, we evaluated the superiority of this model over a conventional orthotopic cell xenograft model. As a result, the novel orthotopic tissue xenograft model reproduced early distant metastasis to the liver and lung, nerve invasion, and cancer-related hypercoagulability of human pancreatic cancer, and showed greater similarity to clinical cases than the control orthotopic cell xenograft model.

The circulating steroid hormones in normal ovulatory cycles accurately control the proliferation and differentiation of the endometrial cells. Androgens, such as testosterone and 5α-dihydrotestosterone, are a type of sex steroid hormones that mainly function in the differentiation, development, and maintenance of male sexual characteristics. Although androgens are best known for their role in male reproduction, the androgen receptor is present in both male and female reproductive organs and is essential for normal reproductive function. Recently, a series of evidence suggests that androgens contribute to endometrial physiology and pathologies. However, the roles of androgens in the endometrium remain insufficiently understood, with contradictory findings being reported. This review summarizes the studies that show the role of androgens in regulating the physiological conditions of the endometrium and the implantation process, and endometrial pathology including endometriosis and others.

Several vesicle-like structures of 100-700 nm diameter flowed onto the fibrous sheets in groups of spindle-shaped bodies arranged in the same direction. Transmission electron microscopy revealed that these structures were encircled by the lipid bilayer membrane. In addition, some had a small mass of high electron density. Scanning electron microscopic observations of specimens treated with bleach revealed particles of almost the same size as the observed electron-dense mass scattered between fibers, with a thickness of approximately 50 nm on day 3 of scale regeneration. The diameter of these particles increased by 5 times on day 14, sticking closely to the fibers. Furthermore, elemental analysis using electron probe microscopy showed that the particles were composed of calcium and phosphorous. These results confirmed that the spindle-shaped bodies and vesicle-like structures were osteoblasts and matrix vesicles, respectively.

It has recently been reported as...Progastrin-releasing peptide (ProGRP), a neuropeptide, is a stable tumor marker for small cell lung carcinoma (SCLC) and other neuroendocrine tumors, such as medullary thyroid carcinoma. It has recently been reported as highly expressed in Ewing sarcoma (ES) and as a useful marker for this type of tumor. However, the mechanisms underlying ProGRP production remain unclear. This study aimed to elucidate its subcellular localization in human ES cell lines. Three ES cell lines (A-673, SK-N-MC, and SK-NEP-1) and one SCLC line (DMS 53) were transplanted into athymic mice. After 3-4 weeks, the tumors were excised, and the expression and localization of ProGRP were examined using immunohistochemistry and immunoelectron microscopy. Immunohistochemistry revealed that all three ES cell lines expressed ProGRP in a dot-like pattern in their cytoplasm, whereas SCLC cells showed a diffuse granular pattern. Immunoelectron microscopy revealed that ProGRP localized in neurosecretory granules (NSGs) in SCLC cells and in autolysosome-like structures in ES cells, with no NSGs observed in the ES cell lines. ES cells secrete high levels of ProGRP, similar to SCLC cells. However, in ES cells, ProGRP is localized in autolysosome-like structures, unlike the NSGs in SCLC, suggesting that ProGRP in ES can be transported and secreted without following the typical pathway for neuropeptides.

This study explored the potential of vi...Ovarian cancer (OvCa) is a leading cause of gynecological cancer-related mortality, primarily due to peritoneal dissemination, which facilitates metastasis in the abdominal cavity. This study explored the potential of vitamin D and its synthetic derivatives in mitigating peritoneal dissemination by modulating the behavior of mesothelial cells (MCs). Vitamin D, through its receptor (VDR), is known to influence cancer progression, and our findings demonstrate that vitamin D derivatives can inhibit mesenchymal transition of MCs induced by TGF-β1, a key driver of peritoneal dissemination. This study used patient-derived primary MCs and in vivo mouse model to assess the effects of vitamin D derivatives on cell morphology, gene expression, and OvCa cell adhesion. Two vitamin D derivatives, VDR agonist, showed significant efficacy in maintaining epithelial-like MC morphology, reducing TGF-β1-induced changes, and inhibiting OvCa cell adhesion to the peritoneum, similar to calcitriol. Conversely, the VDR antagonist derivative induced MC apoptosis, highlighting the essential role of vitamin D in MC survival. These findings suggest that vitamin D derivatives could serve as promising therapeutic agents for OvCa by preserving peritoneal homeostasis and preventing metastasis. Further research is required to explore a broader range of derivatives and their underlying molecular mechanisms.

We report a case of a 70-year...Mucoepidermoid carcinoma in the lung is uncommon, while the occurrence of a collision tumor consisting of primary mucoepidermoid carcinoma (MEC) and typical adenocarcinoma is extremely rare. We report a case of a 70-year-old female with the presence of a nodule in her right lung. The pathological examination revealed a primary collision tumor consisting of invasive adenocarcinoma and mucoepidermoid carcinoma. Manual microdissection was performed to selectively isolate the MEC and adenocarcinoma components, followed by exome sequencing which unveiled identical mutations in both components, suggesting their monoclonal origins with divergent differentiation. Clinical awareness and recognition of such collision tumors are crucial, as they will determine appropriate treatment strategies based on the individual biological aggressiveness of each tumor component.

Pediatric disorders involving the palmar plate of thumb metacarpophalangeal joint include trigger thumb, hyperextension, instability, and dislocation. While anatomical differences exist between children and adults, detailed microstructure evaluations in infants remain unexplored. In this study, we provide a histological and structural assessment of the previously unresolved microstructure of the palmar plate in the thumb metacarpophalangeal joint of infant Japanese macaques (Cercopithecidae, Macaca fuscata), a relevant model for human development. Histological staining (light microscopy) and scanning electron microscopy were employed to visualize the three-dimensional microstructure. The palmar plate of the infant macaque was found to contain (1) elastic fibers, (2) hyaline cartilage composed of type II collagen, and (3) type I collagen fibers arranged in distinct patterns. The cartilaginous region exhibited a reticulate fiber arrangement on its periphery, while the membranous region displayed dense and complex fibers on the proximal phalanx side and parallel on the metacarpal side, respectively. This is the first comprehensive three-dimensional investigation of the infant's thumb's palmar microanatomy, providing a foundation for understanding its development and implications for pediatric disorders.

OSP accounts for about 3-6% of all sinonasal...Oncocytic sinonasal papilloma (OSP) is an uncommon, benign neoplasm characterized by papillary growths arising from the Schneiderian mucosa of the nasal and paranasal sinuses. OSP accounts for about 3-6% of all sinonasal papillomas (SPs), and clinically is often confused with inverted sinonasal papilloma (incidence 73%). Although SP shares some of the clinical characteristics of OSP, the pathogenesis differs and it is very important to distinguish between them. Here we present a case of OSP in the nasal and paranasal sinuses of a 67-year-old Japanese male, who complained of persistent left-sided nasal obstruction. A benign nasal paranasal tumor was diagnosed, and endoscopic sinus surgery was performed. Histopathologically, the tumor cells exhibited an exophytic and endophytic growth pattern, composed of multilayered eosinophilic columnar epithelium with finely granular cytoplasm, and forming microcysts filled with mucin or microabscesses. The epithelium was PTAH-positive, and showed positivity for cytokeratin-7 with oncocytic features. A review of the literature revealed 166 cases of OSP in the nasal and paranasal sinuses, with an overall recurrence rate of about 15%. Here we report a case of OSP in the nasal and paranasal sinuses for which PTAH staining specific for oncocytes was useful, together with a review of the relevant literature.

To ascertain the efficacy of the treatment, pancreatic cancer cells were injected into the thighs of mice and HIFU was applied on one side using continuous waves or trigger pulse waves. Then, tumor volume, tissue changes, and immune marker levels were analyzed. Both the irradiation methods suppressed tumor growth, with the trigger pulse wave showing stronger effects and the difference being significant. Tumor suppression was also observed on the non-irradiated side, suggesting an abscopal effect. These effects vary depending on the irradiation method used. We conclude that HIFU induces both local tumor suppression and a systemic immune response, suggesting its potential for combination with immunotherapy for the treatment of pancreatic cancer.

Herein, we report a case of pagetoid squamous cell proliferation associated with a primary intraosseous carcinoma (PIOC) arising in the periapical lesion of the maxilla, a finding not reported previously. A 60-year-old man presented with prostate adenocarcinoma and bilateral pubic bone, ilium bone, and sacral bone metastases. Radiological examination revealed a cyst that enveloped the apices of the left maxillary first molar roots. Histopathological examination of the cyst specimen indicated squamous cell carcinoma; hence, PIOC was suspected and partial left maxillectomy was performed. Histopathology results showed distant epitheliotrophic spread of atypical clear cells regarding the tumor. The epitheliotrophic cells were positive for cytokeratin (CK)19 and CK7 as odontogenic markers. These phenotypes were similar to those of tumor cells, suggesting pagetoid squamous cell proliferation associated with a PIOC arising. Although diagnosis of this condition is challenging, early detection is vital to ensure prompt treatment and improve patient prognosis.

Quercetin is a natural polyphenolic c...Ankylosing spondylitis (AS) is a chronic inflammatory disease involving the spine and bone joints, which is characterized by hyperosteogeny, ossification of ligaments, and ankylosis. Quercetin is a natural polyphenolic compound with various biological activities such as antioxidant, anti-inflammatory, and anti-tumor. It was to explore the effect of quercetin on AS ossification and its molecular mechanism. In vitro culture of AS mesenchymal stem cells was conducted. Cells were treated with 0, 10, 30, 60, and 80 μM quercetin, divided into control, 10 μM, 30 μM, 60 μM, and 80 μM groups. Alkaline phosphatase (ALP) staining, Alizarin Red staining, real-time quantitative polymerase chain reaction (qRT-PCR), and Western blot (WB) were employed to investigate the effect of quercetin on the expression of osteogenic-related genes and proteins. Additionally, bone morphogenetic protein (BMP) and Smad genes were knocked out to explore quercetin's regulation of BMP/Smad. In vivo experiments were conducted using 50 mice, including 10 in the normal group. An AS model was established in 36 mice, divided into negative control (n = 18, 0.9% saline) and quercetin groups (n = 18, quercetin). Safranin O-fast green (HE) staining and MicroCT scanning were performed before and 4 weeks after injection. In the 60 μM and 80 μM quercetin groups, ALP activity, Ca deposition area, and relative protein/mRNA levels of BMP-1, BMP-2, Smad1, Smad4, and Smad5 in AS mesenchymal stem cells were significantly lower compared to the control, 10 μM, and 30 μM groups (P < 0.05). The 80 μM group exhibited lower levels than the 60 μM group (P < 0.05). In the siRNA + 80 μM group, the reduction in mRNA expression of BMP1, BMP2, Smad1, Smad4, and Smad5 was significantly greater compared to the siRNA group and the 80 μM group (P < 0.05). At 4 weeks post-injection, mice in the quercetin group showed significantly reduced severity of articular cartilage lesions, lymphocyte infiltration, and tissue edema, with no significant increase in sacroiliac joint fusion. Quercetin downregulates the expression of BMP and Smad-related proteins, inhibiting osteogenic differentiation of AS mesenchymal stem cells and effectively reducing ALP activity and Ca deposition levels. These findings suggest that quercetin holds potential application value in the control and treatment of AS disease.

This system is controlled by the extrabacterial environment. The transport routes of the system for VacA have not yet been studied in detail. In this study, we demonstrated by immunoelectron microscopy that VacA localizes closely with the MreB filament in the bacterium, and the MreB polymerization inhibitor A22 obstructs the transport of VacA by ibNoTS. These findings indicate that the route of ibNoTS for VacA is closely associated with the MreB filament Additionally, it was confirmed that VacA does not closely associate with the bacterial filament FtsZ, which is involved in the transport of the virulence factor urease, as previously suggested. We propose that the route of ibNoTS for VacA is associated with the MreB filament in H. pylori.

This study aimed to perform a systemati...Pemphigus vulgaris (PV) is a rare, potentially fatal, immune-mediated chronic disease characterized by the presence of bullous intraepithelial lesions on mucous membranes and skin. This study aimed to perform a systematic literature review covering PV clinical and histopathological aspects and treatment. The literature searches were carried out in the Pubmed, Periódicos Capes, Scopus, Science Direct, Web of Science and Scielo databases. Articles in English or Spanish published from 2000 to 2022 comprising case reports, case series and literature reviews with case report were included. After the analyses, 21 articles were selected. PV generally presents in the third to sixth decades of life and exhibits no gender predilection. The disease manifests itself clinically through irregular and painful blisters that rupture, resulting in erosion and ulceration areas. Histopathologically, the presence of an intraepithelial cleft located above the basal layer and acantholysis are observed. Standard treatment encompasses systemic and topical corticosteroids, with prednisolone being widely employed. Management consists of a remission induction phase and a maintenance phase. An early and accurate diagnosis is paramount to quickly initiate treatment, resulting in more favorable prognoses, as the choice of treatment and responses depend on the severity of the disease. Its expression has been associated with cancer progression and prognosis. Lung cancer has a high incidence rate and poor prognosis. We aim to clarify its expression in lung adenocarcinomas and investigated the utility of SUOX expression as a recurrence factor in operable lung adenocarcinoma. METHODS: We used 60 formalin-fixed paraffin-embedded samples of operable primary lung adenocarcinoma between 2017 and 2018 to immunohistochemically assess SUOX expression levels. Patients were classified into a high or low SUOX expression group, and the associations of SUOX expression with clinicopathological findings and recurrence were analyzed. RESULTS: We revealed that high SUOX expression was significantly (p < 0.05) associated with sex, low Brinkman index, histological type, histological grade and positive for epidermal growth factor receptor (EGFR) mutation. High SUOX expression (HR = 10.218, 95% CI 1.758‒59.376, p = 0.0096) and pathological Stage (HR = 7.538, 95% CI 1.95‒29.14, p = 0.0034) were independently associated with relapse free survival. CONCLUSION: High SUOX expression may be a new indicator of recurrence risk in surgically resected lung adenocarcinomas.

In this case-control study, 54 patients diagnosed with pterygium an...This study aimed to evaluate corneal findings of pterygium cases using in vivo confocal microscopy (IVCM) which is a non-invasive and repeatable method. In this case-control study, 54 patients diagnosed with pterygium and 50 healthy controls were investigated, between 2020 and 2021. After a comprehensive ophthalmological examination, the central corneas of all participants were evaluated by corneal IVCM. Surface epithelial cell, wing cells, basal epithelial cells, nerve fiber density, nerve branch density, anterior and posterior stroma keratocyte densities were measured. Corneas from cases with and without pterygium were similar in terms of age (p = 0.306) and gender (p = 0.564). No statistically significant differences were found in the epithelium cell counts including surface, wing, and basal cells between the two groups (p = 0.950, p = 0.334, and p = 0.624, respectively). No statistically significant differences were observed in subepithelial nerve fiber density (p = 0.849), nerve branch density (p = 0.752), and posterior stromal keratocyte density (p = 0.683) between corneas from cases with and without pterygium. However, corneas from cases with pterygium had significantly higher scores in anterior stromal keratocyte density than those from cases without pterygium (p = 0.045). Corneas from grades 1, 2, and 3 pterygium were similar in cell counts, anterior and posterior stromal keratocyte densities (p > 0.05). However, patients with grade 1 had significantly increased nerve fiber density (p = 0.015) and nerve branch density (p = 0.001) than those with grade 2 and grade 3 pterygium. Corneal IVCM may be useful to better understand the corneal microstructure in cases with pterygium. Comprehensive research on the subject is needed.

This study investigated the therapeutic effect of spinal cord stimulation (SCS) o...Low-extremity ischemic disease is a common complication in diabetic patients, leading to reduced quality of life and potential amputation. This study investigated the therapeutic effect of spinal cord stimulation (SCS) on patients with diabetic foot disease and a rat model of diabetic foot injury. SCS was applied to patients with diabetic foot disease, with clinical assessments performed before and after therapy. Blood levels of NGF, BDNF, and NT-3 were determined by ELISA. A rat model of diabetic foot injury was established to validate NT-3's role in SCS therapy. SCS therapy improved the condition of patients with diabetic ischemic foot disease and promoted wound healing in the rat model. NT-3 levels significantly increased after SCS therapy in both patients and rats. Recombinant NT-3 administration improved wound healing and re-vascularization in the rat model, while NT-3 neutralization abrogated SCS's therapeutic effect. SCS improves the condition of patients with diabetic ischemic foot disease by inducing NT-3 production. Both SCS and NT-3 supplementation show therapeutic potential for ameliorating diabetic foot disease.

Recent research has shown that corneal nerve dysfunction may play a role in certain pathologies of dry eye; however, the details remain unclear. To clarify the aberration in corneal nerves underlying sensory abnormalities, in addition to corneal impairment in dry eye, we examined the morphological alterations of nerve fibers in the corneas excised from guinea pigs with dry eye, where the lacrimal glands were surgically excised. Guinea pigs with dry eye exhibited reduced tear volume, increased spontaneous blink frequency, and corneal epithelial damage. Simultaneously, the subbasal nerve plexus in the cornea visualized using an anti-tubulin βIII antibody partially outgrew and became convoluted. The morphology of peptidergic nerves containing calcitonin gene-related peptide, which may function as a polymodal nociceptor, was also altered. These results indicate that guinea pigs with excised lacrimal glands can serve as useful tools for investigating the neuronal mechanisms underlying corneal pathology in dry eyes. Additionally, chronic tear deficiency may considerably alter nerve structure, including peptidergic nerves in the cornea, accompanied by epithelial damage and increased blink frequency.

We investigated a patient with acute kidney injury with Fanconi syndrome after taking functional food tablets made from red yeast rice using low-vacuum scanning electron microscopy (LVSEM) with an element analysis system. Kidney biopsy revealed proximal tubular necrosis and vacuolization with 10-20 nm black granules, which were similar to the silica nanoparticles found in the functional food tablets and urinary samples, as determined via LVSEM with element analysis. Reabsorbed silica nanoparticles induce oxidative stress in the kidney. Element analysis by LVSEM is useful to investigate a possible cause of acute tubular necrosis in patients with Fanconi syndrome.
